A guitar that I built for a friend has developed a buzz on the 4th string, but only when fretted at the 1st fret. It really sounds like the buzz is coming from the saddle/bridge area, but I've confirmed that the string's ball-end is firmly seated between the bridge plate and bridge pin. Any suggestions?

You might try Putting a fret rocker on the second fret. They always seem to sound like they comming from the bridge.

Very often I've seen kinks in the strings from pressing too hard or being bumpe, or it's a new string that was unwrapped incorrectly. If the kink is right over the fret it will cause noting out.
Changing the string or rubbing the string firmly with a screwdriver handle will fix it it if that's the case.
