I want to fill a humbucker route and the re-finish the body (maple solid body) to appear as the hole was never there. Should I use 100% wood putty or should I glue a piece of wood in the hole, and then fill the remainder with wood putty? idunno

My inclination would be to use wood where possible. The putty might shrink and leave cracks. The wood is stronger.

Wood block, then Bondo (opaque finish, yes?). Wood putty is pretty awful stuff.

Thanks guys, this really helps. I had a lady in Austin that claimed she knew what she was doing work on my bass, and it just didn't turn out the way she said it would. Guess I should have asked one of you guys first. I think I may give it a try.
